Agrobacterium-опосередкована трансформація перспективних генотипів озимої пше­ниці за використання гена орнітин-d-аміно-трансферази

Орнітин-d-амінотрансфераза (ОАТ) є важливим регулятором клітинного ме­таболізму, оскільки реакція, що каталізується цим ферментом, пов’язує кілька біохімічних систем: цикл сечовини, цикл накопичення і деградації проліну та шлях біосинтезу поліамінів. Введення екзогенного гена oat в геном рослин є одним з перспективних методів створення стійких до абіотичних стресів генотипів пшениці. Метою нашої роботи була оптимізація умов Agrobacterium-опосередкованої трансформації морфогенних калюсів нових перспективних генотипів озимої м’якої пшениці та отримання генетично модифікованих рослин з гетерологічним геном орнітин-d-амінотрансферази. Досліджено основні параметри протоколу трансформації, зокрема вплив оптичної щільності агробактеріальної суспензії клітин, концентрації антибіотика цефотаксиму, тривалості кокультивування на частоту отримання канаміциностійких регенерантів з калюсних культур апікального походження. Оптимізовано регенераційне середовище, яке дає змогу прискорити процес отримання генетично модифікованих рослин-регенерантів пшениці та збільшити їх кількість, що забезпечує скорочення біотехнологічного процесу і зменшення матеріальних витрат для його виконання. Шляхом Agrobacterium-опосередкованої трансформації морфогенних калюсів нових перспективних генотипів озимої м’якої пшениці отримано рослини-регенеранти, у геномі яких виявлено повне вбудовування генетичної конструкції, яка містить транс­гени oat та nptII. Трансгенна природа всіх отриманих рослин була підтверджена за допомогою ПЛР з праймерами, специфічними до генів oat та nptII. Частота трансформації для досліджених генотипів становила 0,75—2,5 %.
